Comments: Assay Overview: The purpose of this assay is to determine dose response curves for 128 available out of 143 compounds identified as active in a previously described set of experiments entitled "Primary biochemical high-throughput screening assay for antagonists of the interaction between the Eph receptor B4 (EphB4) and its ligand ephrin-B2 via TNYL-RAW peptide" (PubChem AID = 720). As with the primary assay, this assay is based on the ability of the ligand binding domain of EphB4 (LBD-EphB4) protein to form a complex with the ephrin-B2-antagonistic peptide, TNYL-RAW. TNYL-RAW has been labeled with Alexa-532 dye in order to monitor its interaction with EphB4 by fluorescence polarization. High fluorescence polarization is evidence of receptor-ligand interaction. Low fluorescence polarization reflects the disruption or absence of receptor-ligand interaction.
